Bensulide oxon: In a non-guideline, acute oral cholinesterase study (MRID 50392201), groups of ten adult Sprague-Dawley [Crl:CD(SD)] rats/sex/dose level were administered a single dose of bensulide oxon (98.2% a.i., Lot #: GSO-008) in corn oil via oral gavage at nominal dose levels of 0, 75 (females only), 125, 175, 225, and 275 (males only) mg/kg bw. In addition, groups of ten neonatal pups/sex/dose level were administered a single dose of bensulide oxon via oral gavage dose on post-natal day (PND) 11 at nominal dose levels of 0, 20, 40, 60, and 100 mg/kg bw. Animals were euthanized at the presumed time of peak effect (16 hours for adults; 8 hours for pups). Blood samples were collected prior to euthanasia, and brains were excised, weighed, and submitted for determination of red blood cell (RBC) and brain cholinesterase activities.
No treatment-related effects were observed on mortality, clinical signs of toxicity, body weights, or brain weights in adult rats or PND 11 pups. Although decreased mean brain weights were observed in all adult females (>75 mg/kg bw), this finding was not considered related to treatment because the decreases were not dose dependent, the brain weights were within historical observed ranges, and the decreases were <5% of control values.
RBC cholinesterase activity at the presumed peak time of effect (16 h) was decreased in adult males at ≥125 mg/kg bw and adult females at ≥75 mg/kg bw. Decreases were generally greater with dose in males (↓25-45%) and females (↓25-64%), but the decreases in males appeared to plateau between 175 and 275 mg/kg bw. Brain cholinesterase activity at 16 h was decreased in adult male rats at ≥125 mg/kg bw. The decrease generally was flat, ranging from 8-12%. In adult females, decreased brain cholinesterase activity was observed at ≥175 mg/kg bw compared to respective control values (↓10% and 28%, respectively), with a generally dose-related decrease at 125 mg/kg bw (↓6%).
RBC cholinesterase activity at the presumed peak time of effect (8 h) was decreased in male and female pups at ≥40 mg/kg bw. Decreases were generally greater with dose in males and females (↓24-33% and ↓21-31%, respectively). Brain cholinesterase activity at 8 h also was decreased in male and female pups at ≥40 mg/kg bw. Decreases were generally dose dependent in males and females (↓11-21% and ↓11-26%, respectively).
This study is classified Acceptable/Nonguideline and provided data regarding the effects of a single oral gavage dose of bensulide oxon on brain and RBC cholinesterase inhibition in rat pups and adults.
